Knipps Abstract The prolific multi-drug resistant Gram-negative pathogen Pseudomonas aeruginosa has caused nearly 100,000 human deaths annually in the United States alone. A major reason for this spread of antibiotic-resistant P. aeruginosa is the lack of P. aeruginosa specific antibacterial therapies. The aim of this project was to develop an antibacterial agent that prevents the initial attachment of P. aeruginosa to epithelial cells. We utilized a chimeric protein composed of two domains known to mediate direct bacterial attachment to human cells. The first domain was derived from the bacterial protein FimH and the second from the human protein E-cadherin. FimH binds bacteria to the eukaryotic cell membrane through the aid of the glycosylphosphatidylinositol (GPI) anchor and the GPI-anchored protein E-cadherin is responsible for the initial attachment of the bacteria to the target cell. A FimH-E-cadherin fusion protein was produced and tested for its ability to inhibit the attachment of P. aeruginosa. The levels of inhibition obtained were less than those observed with similar FimH-GPI chimeras tested previously. Taken together, these data suggest that E-cadherin plays a more important role in the attachment process than previously thought.
